چکیده

The chemotherapeutic drug Taxol is known to interact within a specific site on b-tubulin. Although the general location of the site has been defined by photoaffinity labeling and electron crystallography, the original data were insufficient to make an absolute determination of the bound conformation. We have now correlated the crystallographic density with analysis of Taxol conformations and have found the unique solution to be a T-shaped Taxol structure. This T-shaped or butterfly structure is optimized within the b-tubulin site and exhibits functional similarity to a portion of the B9-B10 loop in the a-tubulin subunit. The model provides structural rationalization for a sizeable body of Taxol structure– activity relationship data, including binding affinity, photoaffinity labeling, and acquired mutation in human cancer cells.

The T-Taxol conformation.

T-Taxol is a proposal for the bioactive conformation of paclitaxel (PTX) derived from fitting ligand conformations to the electron crystallographic (EC) density. Although confirmed by a number of studies, some structural ambiguities based on the interpretation of two solid-state REDOR (13)C-(19)F distances in a fluorinated PTX derivative remain. An evaluation of the static and dynamic propertie...

Transformation of taxol-stabilized microtubules into inverted tubulin tubules triggered by a tubulin conformation switch

Bundles of taxol-stabilized microtubules (MTs)--hollow tubules comprised of assembled αβ-tubulin heterodimers--spontaneously assemble above a critical concentration of tetravalent spermine and are stable over long times at room temperature. Here we report that at concentrations of spermine several-fold higher the MT bundles (B(MT)) quickly become unstable and undergo a shape transformation to b...

Characterization of the Taxol Binding Site on the Microtubule IDENTIFICATION OF Arg IN b-TUBULIN AS THE SITE OF PHOTOINCORPORATION OF A 7-BENZOPHENONE ANALOGUE OF TAXOL*

Photoaffinity labeling methods have allowed a definition of the sites of interaction between Taxol and its cellular target, the microtubule, specifically b-tubulin. Our previous studies have indicated that [H]3*-(pazidobenzamido)Taxol photolabels the N-terminal 31 amino acids of b-tubulin (Rao, S., Krauss, N. E., Heerding, J. M., Swindell, C. S., Ringel, I., Orr, G. A., and Horwitz, S. B. (1994...

A Natural Love of Natural Products

Recent research on the chemistry of natural products from the author's group that led to the receipt of the ACS Ernest Guenther Award in the Chemistry of Natural Products is reviewed. REDOR NMR and synthetic studies established the T-taxol conformation as the bioactive tubulin-binding conformation, and these results were confirmed by the synthesis of compounds which clearly owed their activity ...
